目的研究星形胶质细胞划痕损伤后内皮性脂酶(EL)的表达情况。方法取新生大鼠脑皮质,体外分离培养、纯化星形胶质细胞,对其进行划痕损伤,取损伤后不同时间点0 h、1 h、3 h、6 h、12 h、24 h、48 h及正常对照,用Western blot和免疫组化标记检测EL的表达变化及其与凋亡促进因子caspase-3的关系。结果与正常对照组比较,胶质细胞在损伤后6 h EL的表达增高十分明显(P<0.01),在损伤后1h出现caspase-3的表达增高(P<0.01)。结论星形胶质细胞机械性损伤后1 h出现caspase-3表达增高,随后出现EL表达增高,提示星形胶质细胞于损伤后早期即出现凋亡现象,EL在中枢神经系统损伤后修复与再生过程中起重要作用。
Objective To study the expression of endothelial lipase (EL) after astrocyte injury. Methods The cerebral cortex of newborn rats was isolated and cultured. The astrocytes were isolated and cultured in vitro. The astrocytes were harvested and scratched. The injured cells were harvested at 0 h, 1 h, 3 h, 6 h, 12 h, 24 h, 48 h and normal control, Western blot and immunohistochemistry were used to detect the expression of EL and its relationship with apoptosis promoting factor caspase-3. Results Compared with normal control group, the expression of EL in glial cells increased significantly 6 h after injury (P <0.01), and the expression of caspase-3 increased 1 h after injury (P <0.01). Conclusions The expression of caspase-3 is increased at 1 h after mechanical injury of astrocytes, and subsequently the expression of EL is increased. It indicates that astrocytes appear apoptosis in the early stage of injury, and EL repair in the central nervous system after injury Play an important role in the regeneration process.
