目的:研究肿瘤抑制基因um23/NDPK、p53和癌基因p21及胃癌增殖细胞核抗原(PCNA)表达与胃癌浸润、转移的关系。方法:用链霉卵白素-生物素(SAB)免疫组织化学方法对55例经手术切除后的胃癌标本进行um23/NDPK、p53、p21和PCNA的检测。结果:55份胃癌标本中nm23/NDPK、p53、p21、PCNA阳性率分别为71%、29%、38%、84%,其中um23/NDPK、p21及PCNA表达与胃癌浸润深度有关,p21、PCNA阳性和nm23/NDPK表达降低者,癌细胞多已侵入浆膜及装膜外组织;um23/NDPK、p53、PCNA的表达与胃癌的淋巴结转移有关,um23/NDPK表达降低及p53、PCNA阳性者淋巴结转移明显增高(P<0.05)。结论:胃癌组织中存在多种癌基因及抑癌基因表达异常,um23/NDPK的表过降低,p53、p21及PCNA阳性与胃癌浸润范围及淋巴结转移有关系。
Objective: To study the relationship between tumor suppressor genes um23/NDPK, p53 and oncogene p21, and the expression of proliferating cell nuclear antigen (PCNA) in gastric cancer and invasion and metastasis of gastric cancer. Methods: The um23/NDPK, p53, p21 and PCNA were detected in 55 specimens of gastric cancer after surgical resection with streptavidin-biotin (SAB) immunohistochemical method. Results The positive rates of nm23/NDPK, p53, p21 and PCNA in gastric cancer specimens were 71%, 29%, 38% and 84%, respectively. Among them, the expression of um23/NDPK, p21 and PCNA was related to the depth of gastric cancer infiltration. p21, PCNA In patients with positive and nm23/NDPK expression, many cancer cells have invaded serosal and extramural tissues; the expression of um23/NDPK, p53, and PCNA is associated with lymph node metastasis in gastric cancer; the expression of um23/NDPK is decreased; and lymph nodes are positive for p53 and PCNA. The metastasis was significantly higher (P < 0.05). Conclusion: There are many abnormal expressions of oncogenes and tumor suppressor genes in gastric cancer tissues. The expression of um23/NDPK is too low, and the positive rates of p53, p21 and PCNA are related to the invasive range of gastric cancer and lymph node metastasis.
