目的分析高表达天然抗角蛋白自身抗体的情况下,转基因小鼠肾脏的自身免疫状态。方法选择常规条件下饲养的抗角蛋白自身抗体转基因小鼠为研究对象,Bradford法检测尿蛋白并进行聚乙烯酰氨凝胶(SDS-PAGE)电泳;对肾脏组织进行苏木素-伊红(HE)、过碘酸-雪夫(PAS)染色,并行免疫荧光、电镜等分析。结果与同窝对照小鼠相比,抗角蛋白自身抗体转基因小鼠尿蛋白明显升高(P<0.05),以混合性蛋白尿为主:电镜以系膜增殖性肾小球肾炎为主,荧光显微镜下在系膜区可见IgM类物质呈颗粒状沉积(++~+++)。结论抗角蛋白自身抗体转基因小鼠有发生自身免疫性疾病的倾向,自身抗体在肾脏的沉积可能是疾病发生的原因。
OBJECTIVE: To analyze the autoimmune state of kidneys in transgenic mice in the presence of high levels of native anti-keratin autoantibodies. Methods The transgenic mice with anti-keratin autoantibodies raised in routine conditions were selected as the study objects. Urinary protein was detected by Bradford method and subjected to SDS-PAGE electrophoresis. The expression of hematoxylin-eosin (HE) , Periodic acid-Schiff (PAS) staining, parallel immunofluorescence, electron microscopy and other analysis. Results Compared with the littermate control mice, urinary protein of anti-keratin autoantibody transgenic mice was significantly increased (P <0.05), with mixed proteinuria mainly: electron microscopy was mainly mesangial proliferative glomerulonephritis, Under the fluorescence microscope in the mesangial area visible IgM material was granular deposition (++ ~ +++). Conclusion Anti-keratin autoantibodies transgenic mice have a tendency to develop autoimmune diseases. The deposition of autoantibodies in the kidneys may be the cause of the disease.
