目的探讨热休克蛋白70(HSP70)对大鼠脑出血神经细胞凋亡相关基因Bcl-2、Bax蛋白表达的影响。方法将60只SD大鼠分为假手术组,脑出血组各30只,每组又分为6 h、12 h、1 d、2 d、3 d、7 d 6个亚组。假手术组向尾状核注射生理盐水2μL,脑出血组向大鼠尾状核注射2μLⅣ胶原酶,分别于各时间点处死大鼠取脑组织,采用免疫组织化学染色,检测各组大鼠脑出血周围HSP70、Bax、Bcl-2蛋白的表达并进行统计学处理。结果 HSP70、Bcl-2、Bax含量脑出血组较假手术组均明显增高(P<0.01)。HSP70表达在6 h出现,3 d达高峰,7 d表达减少。Bc-2的表达在6 h出现,在1 d时达高峰,第2天维持在高表达状态,随后表达逐渐减少。Bax的表达分别在12 h和3 d达高峰,在1 d时表达呈下降趋势,第7天表达逐渐减少。结论 HSP70高表达对出血脑组织具有明显的保护效应,其促进脑组织抗凋亡蛋白Bcl-2的表达,减少促凋亡蛋白Bax表达,增加Bcl-2/Bax比率,以此抑制脑组织细胞凋亡,从而发挥其对神经细胞的保护作用。
Objective To investigate the effect of heat shock protein 70 (HSP70) on the expression of Bcl-2 and Bax in rat brain after intracerebral hemorrhage. Methods Sixty Sprague-Dawley rats were randomly divided into sham operation group and intracerebral hemorrhage group of 30 rats. Each group was divided into six subgroups: 6 h, 12 h, 1 d, 2 d, 3 d and 7 d. The sham operation group was injected with 2 μL normal saline into the caudate nucleus, and 2 μL Ⅳ collagenase was injected into the caudate nucleus of the cerebral hemorrhage group. The brain tissues were sacrificed at each time point. Immunohistochemistry was used to detect the brain The expression of HSP70, Bax and Bcl-2 protein around the hemorrhage and statistical analysis. Results The levels of HSP70, Bcl-2 and Bax in cerebral hemorrhage group were significantly higher than those in sham operation group (P <0.01). HSP70 expression appeared at 6 h, reaching the peak on the 3rd day and decreased on the 7th day. The expression of Bc-2 appeared at 6 h, peaked at 1 d, remained high at day 2, and then decreased gradually. The expression of Bax peaked at 12 h and 3 d respectively, decreased at 1 d, and decreased at 7 d. Conclusion High expression of HSP70 has a significant protective effect on hemorrhagic brain tissue, which can promote the expression of Bcl-2, decrease the expression of Bax and increase the ratio of Bcl-2 / Bax, and thus inhibit the expression of Bcl- Apoptosis, which play its protective effect on nerve cells.
