目的:研究甘松挥发油对人肾胚胎(HEK)细胞Nav1.5电流的影响。方法:应用细胞膜片钳技术研究甘松挥发油对鼠cDNA编码组成的纯钠通道在人胚胎肾细胞(HEK)表达的Nav1.5细胞离子通道电流的影响。结果:发现甘松挥发油对Na电流的阻滞作用受挥发油浓度影响,量-效反应量呈“S”曲线型,电压钳制影响表现为膜电位越低(电位上移)阻滞作用越强。结论:甘松挥发油对钠电位有明显阻滞作用,其作用随剂量增加而增加,随膜电位降低而作用加强。提示临床可用于因心肌缺血导致膜电位降低而诱发的心律失常。
Objective: To study the effect of volatile oil of Nansong on Nav1.5 current in human renal embryo (HEK) cells. Methods: The cell patch clamp technique was used to study the effect of the volatile oil of Nansong on the ion channel current of pure Na 1 channel in human embryonic kidney (HEK) cells. Results: It was found that the block effect of volatile oil of Nansong on Na current was affected by the concentration of volatile oil, and the amount-response rate was in the curve of “S”. The effect of voltage clamping was that the lower the membrane potential Strong. Conclusion: Nigrum volatile oil has a significant retardation effect on sodium potential, and its role with increasing doses increased, with the role of membrane potential decreased. It is indicated that it can be used clinically for arrhythmia induced by the decrease of membrane potential caused by myocardial ischemia.
