目的:研究芦笋多糖对肿瘤小鼠红细胞离子通道活性的影响。方法:建立了肿瘤动物模型,分高、中、低剂量腹腔给予芦笋多糖7 d,采集并制备红细胞悬液,用试剂盒结合分光光度计测定肿瘤小鼠红细胞膜Na+,K+-ATPase、Ca2+,Mg2+-ATPase活性,采用Fluo-3/AM、MQAE、BCECF-AM荧光探针染色,使用激光共聚焦显微镜和荧光分光光度计分别测定小鼠红细胞内[Ca2+]、[CI-]、[H+]浓度的变化。结果:芦笋多糖能升高肿瘤小鼠红细胞膜Na+,K+-ATPase、Ca2+,Mg2+-ATPase活性,降低肿瘤小鼠红细胞内[Ca2+]浓度,增强红细胞阳离子通道转运活性。芦笋多糖能够降低肿瘤小鼠红细胞内[CI-]浓度,增强红细胞阴离子通道转运活性。芦笋多糖还能够降低肿瘤小鼠红细胞内的[H+]浓度,升高红细胞内pH,维持红细胞酸碱平衡。结论:芦笋多糖通过提高肿瘤小鼠红细胞膜离子通道的功能,稳定细胞内环境,从而有助于保护红细胞膜结构,维持红细胞发挥正常功能。
Objective: To study the effect of asparagus polysaccharide on erythrocyte ion channel activity in tumor mice. Methods: Tumor animal models were established. Asparagus polysaccharide was intraperitoneally administered for 7 days in high, medium and low doses. Erythrocyte suspension was collected and prepared. The erythrocyte membrane Na +, K + -ATPase, Ca2 + Mg2 + -ATPase activity was detected by Fluo-3 / AM, MQAE and BCECF-AM fluorescent probes. [Ca2 +], [CI-] and [H +] were measured by laser confocal microscopy and fluorescence spectrophotometer. Changes in concentration. Results: Asparagus polysaccharide could increase the activity of Na +, K + -ATPase, Ca2 + and Mg2 + -ATPase in the erythrocyte membrane of tumor mice, decrease the concentration of [Ca2 +] in erythrocytes, and enhance the red blood cell cation channel transport activity. Asparagus polysaccharide can reduce the concentration of erythrocyte [CI-] in tumor mice and enhance the transport activity of erythrocyte anion channel. Asparagus polysaccharide can also reduce the concentration of [H +] in erythrocytes of tumor mice, increase the pH of erythrocytes, and maintain the acid-base balance of erythrocytes. Conclusion: Asparagus polysaccharide can improve the membrane structure of erythrocytes and maintain the normal function of erythrocytes by enhancing the function of erythrocyte membrane ion channels in tumor mice and stabilizing the intracellular environment.
