目的:探讨细胞滋养细胞分泌的瘦素(leptin)对自身的生长和侵袭能力的影响。方法:①用RT-PCR及荧光定量PCR方法检测细胞滋养细胞瘦素的表达以及瘦素对其分泌功能的影响。②应用MTT法、体外培养Transwell侵袭模型及运动模型检测瘦素对细胞滋养细胞生长、侵袭及运动能力的影响。结果:①细胞滋养细胞有瘦素mRNA的表达,10ng/ml的瘦素可以显著促进细胞滋养细胞继续分泌瘦素,差异有显著意义(P<0.01);②不同浓度瘦素处理24h及24h后,细胞滋养细胞生长受到抑制,而24h内瘦素可促进滋养细胞侵袭及运动,500ng/ml的瘦素对细胞滋养细胞侵袭及运动的促进作用与对照组比较差异有显著性意义(P<0.05)。结论:细胞滋养细胞分泌的瘦素可以抑制自身的生长,但能增强自身的侵袭及运动。
Objective: To investigate the effect of leptin secreted by cytotrophoblasts on its own growth and invasion. Methods: ① The expression of leptin in cytotrophoblast and the effect of leptin on its secretion were detected by RT-PCR and real-time PCR. ② The effects of leptin on the growth, invasion and exercise capacity of cytotrophoblasts were detected by MTT assay, Transwell invasion model and the model of exercise. Results: (1) The expression of leptin mRNA was found in cytotrophoblast cells. Leptin at 10ng / ml could significantly promote leptin secretion by cytotrophoblast cells (P <0.01); (2) After treated with different concentrations of leptin for 24h and 24h , While the growth of cytotrophoblasts was inhibited. Leptin promoted the invasion and exercise of trophoblasts within 24h, and the effect of 500ng / ml leptin on the invasion and motility of cytotrophoblasts was significantly different from that of the control group (P <0.05 ). Conclusion: Leptin secreted by cytotrophoblast can inhibit its own growth, but can enhance its invasion and exercise.
