目的 研究人脑出血后海马神经元损伤与凋亡促进因子Caspase- 3之间的关系。方法 选取10例因脑出血而死亡的尸检脑标本,应用HE染色,Caspase- 3免疫组织化学染色和Caspase- 3m RNA原位杂交,观察脑出血时海马CA1 区神经元损伤变化与Caspase- 3表达之间的关系。结果 海马CA1 区,出血5 h可检测Caspase- 3免疫组化染色的阳性细胞(10 .4个/高倍视野) ,2 2~4 8h时达高峰(2 1.7~2 4 .3个/高倍视野)。Caspase- 3m RNA原位杂交阳性表达始于5 h(6 .75个/高倍视野) ,2 4 h达高峰(14 .6 0~18.30个/高倍视野) ;二者均在72 h后呈明显下降趋势。出血2 4 h,可检测到TUNEL阳性细胞,持续至72 h。Caspase- 3m RNA与Caspase- 3呈正相关,相关系数为0 .717(P<0 .0 5 )。4~16 h受损神经元形态基本正常,2 4~4 8h细胞凋亡特征明显,72 h后,几乎所有神经元形态呈现严重病理变化。结论 人脑出血后海马神经元发生一系列形态学变化,其演变规律与凋亡促进因子Caspase- 3有密切相关性。
Objective To study the relationship between neuronal damage of hippocampus and apoptosis promoting factor Caspase-3 after intracerebral hemorrhage in rats. Methods Ten autopsy brain samples were collected from cerebral hemorrhage. HE staining, Caspase-3 immunohistochemistry and Caspase-3 mRNA in situ hybridization were used to observe the changes of neuronal damage and the expression of Caspase-3 in hippocampal CA1 area after intracerebral hemorrhage The relationship between. Results In hippocampal CA1 region, the number of Caspase-3 immunohistochemical positive cells (10.4 / high power) was detected at 5 h after the hemorrhage, reaching the peak at 2 ~ 48 h (2 1.7 ~ 2.4 3 high power fields ). The positive expression of Caspase-3 mRNA in situ hybridization began at 5 h (6.75 high power fields) and peaked at 14 h (14.60 to 18.30 high power fields), both of which were obvious at 72 h Downtrend. Twenty-four hours after bleed, TUNEL-positive cells could be detected for up to 72 h. Caspase-3 mRNA was positively correlated with Caspase-3 with a correlation coefficient of 0.717 (P <0.05). The morphous of injured neurons was basically normal at 4 ~ 16 h, and the apoptotic features were obvious at 24 h to 48 h. After 72 h, almost all the neurons showed serious pathological changes. Conclusion A series of morphological changes of hippocampal neurons occur after intracerebral hemorrhage. The evolution of them is closely related to the expression of Caspase-3.
