替米沙坦对自发性高血压大鼠颈动脉Ⅰ型、Ⅲ型胶原及转化生长因子β1表达的影响

目的:研究自发性高血压大鼠(SHR)颈动脉Ⅰ型、Ⅲ型胶原及转化生长因子1β(TGF-1β)的表达,并探讨替米沙坦的干预作用。方法:30只雄性12周龄的SHR大鼠随机分为3组:高血压组(SHR)、替米沙坦低剂量组(T e lL)、替米沙坦高剂量组(T e lH),并设同周龄雄性的W KY大鼠为对照组(W KY,n=10),干预18周。观察各组大鼠收缩压(SBP)、免疫组化评估颈动脉Ⅰ型、Ⅲ型胶原及TGF-1β的表达。结果:(1)2周后T e lH组SBP明显低于SHR组(P<0.01),其降压作用持续至实验结束,而T e lL组SBP与SHR组差异无统计学意义(P>0.05);(2)SHR组颈动脉外膜Ⅰ型胶原的表达明显高于W KY组(P<0.01),T e lH、T e lL组颈动脉外膜Ⅰ型胶原的表达低于SHR组(P<0.05);(3)SHR组颈动脉外膜Ⅲ型胶原的表达明显低于W KY组(P<0.01),T e lH组颈动脉外膜Ⅲ型胶原的表达高于SHR组(P<0.01);(4)SHR组颈动脉中膜TGF-1β的IOD值明显低于W KY组(P<0.01),T e lH、T e lL组颈动脉中膜TGF-1β的IOD值高于SHR组(P<0.05)。结论:SHR颈动脉外膜Ⅰ型胶原表达增多,Ⅲ型胶原表达减少,替米沙坦能减少颈动脉外膜Ⅰ型胶原表达、增多Ⅲ型胶原表达,其减少颈动脉外膜Ⅰ型胶原表达的机制可能与替米沙坦增加中膜TGF-1β表达有关。 Objective: To investigate the expression of type Ⅰ, type Ⅲ collagen and transforming growth factor-1β (TGF-β1) in spontaneously hypertensive rats (SHR) and to explore the intervention effect of telmisartan. Methods Thirty male SHR rats of 12 weeks old were randomly divided into three groups: hypertension group (SHR), telmisartan low dose group (T e LL), telmisartan high dose group (T e lH) , And W KY rats of the same age were set as the control group (W KY, n = 10) for 18 weeks. The systolic blood pressure (SBP), the expression of type Ⅰ, type Ⅲ collagen and TGF-β1 in the carotid arteries were observed by immunohistochemistry. Results: (1) After 2 weeks, the SBP in T e lH group was significantly lower than that in SHR group (P <0.01), and its antihypertensive effect continued to the end of the experiment, but there was no significant difference between T SB and SHR group (P> 0.05). (2) The expression of type I collagen in carotid artery adventitia of SHR group was significantly higher than that in W KY group (P <0.01). The expression of type I collagen of carotid adventitia in SHE group was lower than that in SHR group (P <0.05). (3) The expression of type Ⅲ collagen of carotid adventitia in SHR group was significantly lower than that in W KY group (P <0.01). The expression of type Ⅲ collagen in carotid adventitia of T e lH group was higher than that in SHR group P <0.01). (4) The IOD value of TGF-β in the carotid artery of SHR group was significantly lower than that in W KY group (P <0.01). The IOD value of TGF- Higher than SHR group (P <0.05). Conclusion: The expression of type Ⅰ collagen in the carotid artery of SHR increased and the expression of type Ⅲ collagen decreased. Telmisartan can reduce the expression of type Ⅰ collagen in the carotid adventitia, increase the expression of type Ⅲ collagen and decrease the expression of type Ⅰ collagen in the carotid artery adventitia May be related to the mechanism of telmisartan increasing the expression of mesangial TGF-1β.
