目的通过体外实验研究3种可用于慢性肝炎患者的注射液对4种CYPs亚型酶活性的影响。方法应用体外“cocktail”反应体系,利用LC-MS/MS法测定代谢产物,检测与异甘草酸镁、利巴韦林及与胸腺五肽注射液共孵育后,CYP2C9、2C19、2D6与3A4的体外酶活性。结果在体外“cocktail”实验中,以上3种注射液对于CYP2C9、2C19、2D6与3A4的体外酶活性均未显示出明显的抑制作用(P>0.05)。结论异甘草酸镁、利巴韦林及胸腺五肽注射液与CYP2C9、2C19、2D6与3A4的底物发生具有临床意义的药物相互作用的可能性较小。
Objective To study the effects of three injections of CYP3A1 and CYP1A1 on the CYP3A1 CYP2A1 activity in chronic hepatitis patients in vitro. Methods The metabolites were determined by LC-MS / MS method in vitro with “cocktail” reaction system. After incubation with magnesium isoglycyrrhizinate, ribavirin and thymopentin injection, CYP2C9, 2C19, 2D6 and 3A4 were tested in vitro Enzyme activity. Results In vitro “cocktail” experiments, the above three injections did not show any significant inhibitory effect on the in vitro enzymatic activity of CYP2C9, 2C19, 2D6 and 3A4 (P> 0.05). CONCLUSIONS: It is less likely that clinically significant drug-drug interactions will occur with isoglycyrrhizinate, ribavirin, and thymopentin injections and CYP2C9, 2C19, 2D6 and 3A4 substrates.
