The decline of human gastric cancer BGC-823 cells’ proliferation and the responsiveness to epidermal growth factor, following down regulation of c-erbB-2 expression by the antisense recombinant vector, was observed . The induction of cell proliferation, c-myc gene expression and expression of EGF receptor gene by EGF were also inhibited obviously, while the expression of the cyclin dependent kinase inhibitor (p21) was significantly enhanced. The analysis of the activation mechanism of p21 transcription revealed that STAT1 bound specifically with the promoter of p21 gene then accelerated transcription of the gene.
