干旱是限制黄芪种子自然萌发的直接因素。在PEG-6000溶液中模拟干旱胁迫环境,采用综合抗旱系数、权重隶属函数值、主成分分析、聚类分析等方法相结合,利用农艺性状指标,对12个种源黄芪种子萌发期进行抗旱性鉴定,结果表明:综合抗旱系数和权重隶属函数值均以LQ1(陇芪1号)、WC(武川)、LQ2(陇芪2号)种源较高。种子抗旱性的主成分分为三个,第Ⅰ主成分为种子萌发因子,包括发芽指数、发芽率、发芽势和活力指数;第Ⅱ主成分为种苗生长因子,包括胚根长、苗总长、胚芽长;第Ⅲ主成分为种苗健康因子,包括苗干重和根芽比。三个主成分呈现层层递进关系,各指标间具有相关关系的占44.44%。聚类分析将12个黄芪种源分为5类,WC、LQ12个种源抗旱指标度量值最大,聚为一类;TY(太原)的抗旱性最低,单独聚为一类。采用多种方法综合分析黄芪种子萌发期抗旱性,能够从个体、群体、整体三个不同层次准确评价其抗旱性,为黄芪种质资源利用、引种和育种提供科学依据。
Drought is a direct factor limiting the natural germination of Astragalus seed. The drought stress environment was simulated in PEG-6000 solution. The drought resistance index, the membership function value, the principal component analysis and cluster analysis were used to analyze the drought resistance of 12 provenances The results showed that both the comprehensive drought resistance coefficient and the weight membership function value were higher in LQ1 (Longqiu1), WC (Wuchuan) and LQ2 (Longqiu2). The main components of seed drought resistance were divided into three parts, the main component of the first seed germination factor, including germination index, germination rate, germination potential and vigor index; the second main component of seedling growth factor, including radicle length, total length of seedlings , Germ length; the third main component of seedling health factors, including seedling weight and root shoot ratio. The three principal components showed a progressive relationship with each other, accounting for 44.44% of the total. Cluster analysis divided 12 Astragalus species into five categories, and the drought and drought index of WC and LQ12 provenances were the largest and clustered into one category. TY (Taiyuan) had the lowest drought resistance and clustered together into one category. Comprehensive analysis of the drought resistance of astragalus seed germination period using a variety of methods can accurately evaluate the drought resistance of Astragalus from three different levels of individuals, populations and provide a scientific basis for the utilization, introduction and breeding of astragalus germplasm resources.
