目的运用原子力显微镜(Atomic Force Microscope,AFM)观察牙周炎患者龈下优势菌表面超微形态特征,分析糖尿病牙周炎、非糖尿病牙周炎研究对象菌斑中优势菌表面形态是否存在差异,探讨有致病作用的优势菌形态与两种类型牙周炎病理损伤之间的相关性。方法选取糖尿病牙周炎、非糖尿病牙周炎典型病例各30例,应用AFM对符合设定标准的优势菌进行扫描。测量每个菌体表面平均粗糙度、峰值、隆起平均大小,观察其纳米结构变化,评价两种类型牙周炎龈下优势菌的形态学特征。结果通过研究发现糖尿病牙周炎组、非糖尿病牙周炎组龈下优势菌表面差异存在统计学意义,糖尿病牙周炎组优势菌表面的平均粗糙度、峰值、隆起平均大小均明显高于非糖尿病牙周炎组(P<0.05)。结论糖尿病牙周炎龈下优势菌表面的纳米级超微形态不同于非糖尿病牙周炎,其差异性显示两种类型牙周炎的牙周损伤特点与其各自龈下优势菌的形态结构相关。
Objective To observe the ultrastructural features of subgingival dominant bacteria in patients with periodontitis using Atomic Force Microscope (AFM), and to analyze whether the surface morphology of dominant bacteria in plaque of diabetic periodontitis and non-diabetic periodontitis is different, To investigate the correlation between pathogenic lesions and pathological lesions of two types of periodontitis. Methods A total of 30 typical cases of periodontitis and non-diabetic periodontitis were selected, and AFM was used to scan the dominant bacteria that meet the set criteria. The average surface roughness, peak value and bulge of each cell surface were measured. The changes of the nanostructures were observed. The morphological characteristics of the subgingival dominant bacteria of the two types of periodontitis were evaluated. Results The results showed that there was a significant difference in the surface of subgingival predominant bacteria in diabetic periodontitis group and non-diabetic periodontitis group. The mean roughness, peak value and mean bump of dominant bacteria in diabetic periodontitis group were significantly higher than those in non-diabetic periodontitis group Diabetic periodontitis group (P <0.05). Conclusions The nanoscale ultrastructure of subgingival dominant bacteria on diabetic periodontitis is different from that of non-diabetic periodontitis. The difference between the two groups shows that the periodontal injury characteristics of the two types of periodontitis are related to the morphological structure of their respective subgingival dominant bacteria.
