目的研究3-正丁基苯酞(NBP)在大鼠体内的组织分布。方法采用Hypersil ODS2色谱柱(4.6 mm×250 mm,5μm),流动相为0.05 moL.L-1醋酸钠溶液-乙睛(50∶50,用醋酸调节pH 4.5),检测波长228 nm,柱温40℃。大鼠按3-正丁基苯酞混悬液单剂量360 mg.kg-1灌胃给药,采用高效液相色谱法测定各组织中3-正丁基苯酞的含量。结果 3-正丁基苯酞在各组织样品中的线性范围均为0.049 8~9.96μg.mL-1,线性关系良好。给药后1 h在肾中3-正丁基苯酞的浓度最高,其次是脑、心和肝。结论该方法灵敏度高,专属性强,结果准确可靠,适合用于研究3-正丁基苯酞在大鼠中的组织分布。研究表明,3-正丁基苯酞在大鼠体内分布快而广,且可通过血脑屏障。
Objective To study the tissue distribution of 3-n-butylphthalide (NBP) in rats. Methods Hypersil ODS2 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase was 0.05 mol L -1 sodium acetate solution (50:50, pH 4.5 with acetic acid). The detection wavelength was set at 228 nm and the column temperature 40 ° C. Rats by 3-n-butylphthalide suspension single dose of 360 mg.kg-1 intragastric administration, using high performance liquid chromatography determination of 3-n-butylphthalide in each tissue content. Results The linear range of 3-n-butylphthalide was 0.049 8 ~ 9.96μg.mL-1 in all tissue samples with good linearity. The concentration of 3-n-butylphthalide in the kidney was highest at 1 h after administration, followed by brain, heart and liver. Conclusion The method is sensitive, specific and accurate. It is suitable for studying the tissue distribution of 3-n-butylphthalide in rats. Studies have shown that 3-n-butylphthalide distribution in rats fast and wide, and can cross the blood-brain barrier.
