目的建立体外培养的人小梁(human trabecular meshwork,HTM)细胞,并观察小梁细胞细胞骨架与细胞连接的形态学特征。方法体外培养的HTM细胞传代后,免疫组化方法鉴定,电镜观察小梁细胞的超微结构,激光共聚焦显微镜观察actin、vinculin、β-catenin的表达。结果通过光镜、透射电镜、扫描电镜和免疫组化的方法证实体外培养的细胞为人眼小梁细胞。人眼小梁细胞的细胞骨架与细胞连接丰富。结论体外培养的小梁细胞细胞骨架结构明显,证明该细胞有收缩能力;β-连接素与纽蛋白着染明显,说明体外培养的小梁细胞,其细胞与细胞间、细胞与细胞基质间的连接仍具有活体小梁细胞的特性。
Objective To establish human trabecular meshwork (HTM) cells cultured in vitro and observe the morphological characteristics of the trabecular meshwork cells. Methods HTM cells cultured in vitro were passaged and identified by immunohistochemistry. The ultrastructure of trabecular meshwork cells was observed by electron microscope. The expression of actin, vinculin and β-catenin was observed by laser scanning confocal microscopy. Results The cells cultured in vitro were identified as human eye trabecular cells by light microscopy, transmission electron microscopy, scanning electron microscopy and immunohistochemistry. Human eye trabecular meshwork cells and cellular cytoskeletal rich. CONCLUSION: The cytoskeleton of trabecular cells cultured in vitro is obvious, which shows that the cells have contractile ability. Β-catenin and vinculin stain obviously, indicating that trabecular cells cultivated in vitro, their cells and cells, between cells and the cell matrix The connection still has the characteristics of a living trabecular cell.
