目的:研究芍药软肝合剂的清除自由基、抗氧化能力和对肝细胞损伤的保护作用,从抗氧化应激角度探讨其临床治疗化疗药物性肝损伤的作用和机制。方法:采用DPPH、羟自由基和超氧阴离子自由基考察芍药软肝合剂的清除自由基作用。采用总抗氧化能力、脂质过氧化水平和亚铁离子络合能力考察芍药软肝合剂的抗氧化作用。采用过氧化氢诱导的肝细胞L-O2损伤模型评价芍药软肝合剂对肝损伤的保护作用。结果:芍药软肝合剂对DPPH自由基、羟基自由基以及超氧阴离子自由基均具有较强的清除能力,还具有较强的总抗氧化能力,抗脂质过氧化能力和金属离子络合能力。对过氧化氢引起的肝细胞死亡也就具有较好的抑制作用。结论:芍药软肝合剂的抗氧化应激能力可能是其临床其治疗化疗药物性肝损伤的的机制之一。
OBJECTIVE: To study the scavenging free radical, antioxidant capacity and protective effect of Shaoyao Ruangan Mixture on hepatocellular injury, and to explore its clinical effect and mechanism of chemotherapeutic drug-induced liver injury from the perspective of antioxidant stress. Methods: Scavenging free radicals of Paoyu Ruangan Mixture were investigated by DPPH, hydroxyl radical and superoxide anion free radical. The anti-oxidative effect of Shaoyu Ruan-He Mixture was investigated by total antioxidant capacity, lipid peroxidation and ferrous ion complexation ability. Hydrogen peroxide-induced L-O2 injury model of hepatocytes was used to evaluate the protective effect of Shaoyu Ruan Gan Mixture on liver injury. Results: Shaoyao Ruangan Mixture has strong scavenging ability on DPPH free radical, hydroxyl free radical and superoxide anion free radical. It also has strong total antioxidant capacity, anti-lipid peroxidation ability and metal ion complexing ability . Hydrogen peroxide caused by liver cell death also has a better inhibitory effect. Conclusion: The antioxidant capacity of Shaoyu Ruanan Mixture may be one of its mechanisms in the treatment of chemotherapy-induced liver injury.
