目的:探讨子痫前期产妇胎盘组织中基质金属蛋白酶-3(MMP-3)及其抑制剂-3(TIMP-3)表达及可能发挥的作用。方法:采用子痫前期患者(子痫前期组)及正常妊娠孕妇(对照组)胎盘组织,采用免疫组化法检测MMP-3和TIMP-3水平,以HE染色观察绒毛血管密度。结果:子痫前期组与对照组胎盘组织中MMP-3和TIMP-3均有不同程度的阳性表达,其在对照组的绒毛滋养细胞中表达高于蜕膜组织(P<0.05),子痫前期组在绒毛滋养细胞与蜕膜组织中表达程度接近,子痫前期组无论是绒毛滋养细胞中还是蜕膜组织中阳性表达均高于对照组(P<0.05);镜检子痫前期胎盘组织绒毛血管密度明显低于对照组(P<0.05)。结论:子痫前期患者与正常妊娠孕妇比较胎盘组织中MMP-3表达下调而TIMP-3的表达上调,提示子痫前期的发病及发生可能与MMP-3和TIMP-3的表达异常表达有关。
Objective: To explore the possible role of matrix metalloproteinase-3 (MMP-3) and its inhibitor-3 (TIMP-3) in the placenta of preeclampsia. Methods: The placental tissues of preeclampsia (preeclampsia group) and normal pregnant women (control group) were collected. The levels of MMP-3 and TIMP-3 were detected by immunohistochemistry and the villus density was observed by HE staining. Results: The levels of MMP-3 and TIMP-3 in placenta of preeclampsia and control group were all significantly higher than those in control group (P <0.05), eclampsia The expression level in villous trophoblasts and decidua tissues in pre-eclampsia group was close to that in pre-eclampsia group, either in villous trophoblast cells or in decidua tissues (P <0.05). The levels of placental tissue in preeclampsia Villus vascular density was significantly lower than the control group (P <0.05). Conclusion: Compared with normal pregnant women, the expression of MMP-3 and MMP-3 in placenta of pregnant women with preeclampsia are up-regulated, suggesting that the pathogenesis of preeclampsia may be related to the abnormal expression of MMP-3 and TIMP-3.
