目的探讨钙激活性钾通道(KCa)在缓激肽(bradykinin,BK)选择性开放血脑肿瘤屏障(blood-braintumor barrier,BTB)中的作用。方法建立脑胶质瘤大鼠模型,颈内动脉灌注BK后采用免疫组化ABC法和Westernblot法测定肿瘤组织KCa通道蛋白的分布和含量的变化,以t检验进行统计分析。结果脑胶质瘤大鼠模型经颈内动脉灌注BK后,其肿瘤内血管内皮细胞的KCa通道蛋白增加,且以灌注后10 min增加最为显著。结论KCa通道的表达增多是BK选择性开放BTB机制中的一个重要环节。
Objective To investigate the role of calcium-activated potassium channel (KCa) in the selective opening of blood-brain tumor barrier (BTB) by bradykinin (BK). Methods The rat model of glioma was established. After BK was injected into internal carotid artery, the distribution and content of KCa channel protein in tumor tissue were determined by immunohistochemical ABC and Western blot. The statistical analysis was performed by t test. Results The rat model of glioma was infused with BK via the internal carotid artery, which increased the number of KCa channel proteins in the endothelial cells of the tumor and increased most significantly at 10 min after perfusion. Conclusion The increased expression of KCa channel is an important link in BK selective open BTB mechanism.
