目的:观察复方瑞康欣对吗啡依赖的SK-N-SH细胞各型NOS活性和NO浓度的影响并探讨其机制。方法:SK-N-SH细胞周期同步化后,用吗啡(10μmol/L)作用48h,然后用复方瑞康欣含药血清干预12h,再用纳洛酮(10μmol/L)作用10min后,检测NOS活性和NO含量。结果:复方瑞康欣含药血清能抑制总NOS和cNOS活性(P<0.01),从而降低NO含量(P<0.01),但对iNOS活性没有影响(P>0.05)。结论:复方瑞康欣的戒毒作用与NOS活性和NO水平有关。
Objective: To observe the effect of Fufang Ruikangxin on NOS activity and NO concentration in various types of morphine-dependent SK-N-SH cells and to explore its mechanism. Methods: After synchrocytes of SK-N-SH cells were treated with morphine (10μmol / L) for 48h, the cells were treated with Ruikangxin serum for 12h and then treated with naloxone (10μmol / L) for 10min. NOS activity and NO content. Results: The drug-containing serum of Ruikangxin inhibited the activity of total NOS and cNOS (P <0.01), reduced the content of NO (P <0.01), but had no effect on iNOS activity (P> 0.05). Conclusion: The drug rehabilitation of compound Ruikangxin is related to NOS activity and NO level.
