目的研究三氯乙烯(TCE)对大鼠睾丸细胞DNA损伤和睾丸组织氧化应激的作用。方法选用雄性SD大鼠,分为3组,每组5只,用1500、3000 mg/kg的TCE经口灌胃染毒,48 h处死动物。用单细胞凝胶电泳(彗星实验)检测睾丸细胞DNA损伤并检测睾丸组织中活性氧(ROS)、丙二醛(MDA)、还原型谷胱甘肽(GSH)的含量。结果TCE在1 500、3000 mg/kg剂量条件下,睾丸细胞DNA的Olive尾矩增加,DNA损伤细胞率也明显增高(P<0.01);大鼠睾丸组织的ROS、MDA均较对照组显著增高(P<0.01),同时还原型GSH又较对照组明显下降(P<0.01)。结论TCE在1 500、3000 mg/kg剂量条件下染毒大鼠后,造成睾丸组织明显的氧化应激并引起睾丸细胞DNA损伤。
Objective To investigate the effect of trichlorethylene (TCE) on DNA damage and oxidative stress in testicular tissue of rats. Methods Male Sprague-Dawley rats were randomly divided into 3 groups with 5 rats in each group. Animals were administered with 1500,3000 mg / kg TCE via oral gavage and sacrificed 48 h later. The DNA damage of testicular cells was detected by single cell gel electrophoresis (comet assay) and the contents of reactive oxygen species (ROS), malondialdehyde (MDA) and reduced glutathione (GSH) were determined. Results At the dose of 1 500 and 3000 mg / kg TCE, the Olive tail moment of testis DNA increased and the DNA damage rate was significantly increased (P <0.01). The ROS and MDA in testis of rats were significantly higher than those in control group (P <0.01), at the same time reducing GSH was significantly lower than the control group (P <0.01). Conclusion TCE exposure to 1 500 and 3000 mg / kg of rat resulted in significant oxidative stress in testis and DNA damage in testicular cells.
