目的探讨人口腔鳞癌体外培养亲本细胞株KB与耐药细胞株KBV200的蛋白质表达差异。方法在体外培养KB及KBV200细胞株,采用表面增强激光解析电离飞行时间质谱技术(SELDI)及蛋白质芯片技术检测KB及KBV200蛋白质谱。结果体外培养的口腔鳞癌亲本细胞株与耐药细胞株的蛋白质存在差异表达,CM-10芯片共捕获102个蛋白,发现差异峰19个,其中15个差异蛋白在KBV200细胞中高表达,4个差异蛋白在KBV200细胞中低表达。结论SELDI蛋白芯片技术检测口腔鳞癌亲本细胞株与耐药细胞株蛋白质的差异表达方法简便、敏感、重复性好。
OBJECTIVE: To investigate the differences of protein expression between KB cells and KBV200 cells cultured in vitro. Methods KB and KBV200 cell lines were cultured in vitro. The protein profiles of KB and KBV200 were detected by surface enhanced laser desorption / ionization time of flight mass spectrometry (SELDI) and protein chip technology. Results The proteins of oral squamous cell carcinoma cell lines and drug-resistant cell lines were differentially expressed in CM-10 cells. A total of 102 proteins were found in the CM-10 chip, and 19 differentially expressed proteins were found. Among them, 15 differentially expressed proteins were highly expressed in KBV200 cells and 4 Differential proteins were low expressed in KBV200 cells. Conclusion SELDI protein chip technology to detect oral squamous cell carcinoma cell lines and resistant cell lines protein differential expression method is simple, sensitive and reproducible.
