目的了解陕西省麻疹病毒流行株的基因型别,探讨控制麻疹策略。方法在2000~2003年采集陕西省麻疹爆发和散发病人的咽喉拭子标本56份,用EB病毒转化的狨猴淋巴母细胞(B95a)分离到麻疹病毒15株。用逆转录-聚合酶链反应(RT-PCR)从7株麻疹病毒中扩增出核蛋白(N)基因羧基端450个核苷酸片段,进行核苷酸序列测定和基因分型。结果7株麻疹病毒均属H1基因型,其中H1a4株,H1b3株,H1a和H1b在450个核苷酸片段中的差异为2%~4%。结论H1a和H1b在陕西省隔年流行,流行优势株的变化可能受本土病毒变异和邻省输入病毒的双重影响。
Objective To understand the genotypes of measles virus in Shaanxi Province and to explore the strategy of measles control. Methods A total of 56 throat swabs were collected from patients in Shaanxi Province during 2000-2003. Fifteen measles viruses were isolated from the EB virus-transformed marmoset lymphoblasts (B95a). Nucleotide 450 nucleotides fragment of nucleoprotein (N) gene was amplified by RT-PCR from seven measles viruses, and the nucleotide sequence and genotyping were performed. Results All seven measles viruses belonged to the H1 genotype. Among them, H1a4, H1b3, H1a and H1b showed a difference of 2% ~ 4% in 450 nucleotide fragments. Conclusions H1a and H1b are prevalent in Shaanxi Province in the next year. The changes of prevailing strains may be affected by the variation of indigenous viruses and the imported viruses of neighboring provinces.
