目的研究氟中毒大鼠肝组织中caspase-3、caspase-9 mRNA表达的变化。方法将48只健康清洁级SD雄性大鼠随机分为4组,分别为对照(去离子水)组和低(50 mg/L)、中(100 mg/L)、高剂量(200 mg/L)氟染毒组,每组12只。采用自由饮水方式进行染毒,连续染毒120 d。采用荧光定量PCR法检测肝组织中caspase-3、caspase-9 mRNA的表达水平。结果与对照组相比,高剂量氟染毒组caspase-3、caspase-9 mRNA的表达水平明显升高,差异均有统计学意义(P<0.05)。结论过量氟从基因水平促进大鼠肝脏组织中caspase-9、caspase-3的表达。
Objective To study the changes of caspase-3 and caspase-9 mRNA expression in liver tissues of fluorosis rats. Methods Forty - eight healthy male SD rats were randomly divided into 4 groups: control (deionized water) group and low (50 mg / L), medium (100 mg / L) Fluoride exposure group, 12 rats in each group. Adopt free drinking way to carry on poisoning, continuous poisoning 120 d. Quantitative real-time PCR was used to detect the expression of caspase-3 and caspase-9 mRNA in liver tissue. Results Compared with the control group, the expression of caspase-3 and caspase-9 mRNA in high-dose fluorosis group were significantly increased, the difference was statistically significant (P <0.05). Conclusion Excessive fluoride can promote the expression of caspase-9 and caspase-3 in rat liver tissue at gene level.
