目的:观察熄风定颤汤(以下简称中药)对PD模型大鼠旋转行为和纹状体单氨类神经递质的影响,探讨其可能作用机制。方法:采用6-OHDA偏侧纹状体两点注入法制备PD模型大鼠,随机分为模型组、美多巴组、中药高、中、低剂量组,并设假手术组为对照组,灌胃治疗4周。观察各组大鼠治疗前后APO诱导的行为学变化及对纹状体单胺类神经递质的影响。结果:美多巴组灌胃后2周旋转行为有所下降(P<0.01),但4周后又有所增加(P>0.05);中药高、低剂量组灌胃后4周后旋转行为有明显改善(P<0.05,P<0.05);中药中剂量组灌胃后2周、4周后旋转行为均有明显改善(P<0.05,P<0.01)。美多巴组、中药高、中、低剂量组大鼠损毁侧纹状体DA含量、HVA、DOPAC含量均明显增加,其中中剂量组增加最明显,分别比损毁对侧下降了21.78%、23.53%、3.52%,与损毁对侧、假手术组损毁侧差异不显著(P均>0.05),与模型组大鼠DA和DOPAC相比差异显著(P<0.01,P<0.05)。结论:熄风定颤汤可明显改善PD模型大鼠的旋转行为、增加纹状体内DA及其代谢产物的含量,以中剂量组效果最明显。
Objective: To observe the effect of Xifeng Dingchan Decoction (hereinafter referred to as TCM) on the rotation behavior and monoamine neurotransmitter in striatum of rats with PD model and to explore its possible mechanism. Methods: PD model rats were prepared by injecting 6-OHDA into lateral striatum. The rats were randomly divided into model group, metoprol group, high, medium and low dose Chinese herbal medicine group, sham-operation group as control group, Gavage for 4 weeks. The changes of APO-induced behavioral changes and the effects on monoamine neurotransmitters in striatum were observed before and after treatment in each group. Results: The rotational behavior of the two groups decreased after 2 weeks of administration (P <0.01), but increased after 4 weeks (P> 0.05). After 4 weeks of administration, the rotation behavior (P <0.05, P <0.05). The rotational behavior of the middle-dose group was significantly improved after 2 and 4 weeks of treatment (P <0.05, P <0.01). The contents of DA, HVA and DOPAC in the injured group were significantly increased in the group of the meduoba and the group of medium, medium and low dose of traditional Chinese medicine, the most obvious increase was in the medium dose group, which decreased by 21.78% and 23.53 % And 3.52% respectively. There was no significant difference between the two groups (P> 0.05). The difference was significant between DA and DOPAC in model group (P <0.01, P <0.05). Conclusion: Xifeng Dingchan Decoction can obviously improve the rotation behavior of rats with PD and increase the contents of DA and its metabolites in the striatum, and the effect is most obvious in the middle dose group.
