目的:探讨递增大强度训练对大鼠骨骼肌PI3K/Akt信号转导通路的影响。方法:12只雄性SD大鼠随机分为安静组和递增大强度运动组,每组6只。运动组进行递增大强度跑台训练。8周后采用Real-time PCR和Western blotting等测定大鼠比目鱼肌和腓肠肌Akt(v-akt murine thymoma viral oncogene homolog)、PTEN(phosphatase and tensin homologue)和磷脂酰肌醇-3-激酶(phosphatidylinositol-3-kinase,PI3K)的mRNA相对表达量和蛋白表达水平。结果:(1)与安静组比较,递增大强度运动组比目鱼肌和腓肠肌Akt mRNA相对表达量显著下调,而腓肠肌Akt蛋白表达显著上调;(2)递增大强度运动组比目鱼肌和腓肠肌PTEN mRNA及蛋白表达均无显著变化;(3)递增大强度运动组比目鱼肌PI3K蛋白表达显著上调,比目鱼肌和腓肠肌PI3K mRNA相对表达量无显著变化。结果提示:Akt mRNA下调的途径可以独立于PI3K之外,且递增大强度跑台训练可以诱导这种特异Akt失活途径的启动;在激活Akt的方式上除了通过PI3K以外,尚有其他的激活事件发生。
OBJECTIVE: To investigate the effect of increasing intensity training on the PI3K / Akt signal transduction pathway in skeletal muscle of rats. Methods: Twelve male Sprague-Dawley rats were randomly divided into two groups: sedentary group and high-intensity exercise group. Exercise group to increase intensity treadmill training. After 8 weeks, the expressions of Akt (v-akt murine thymoma viral oncogene homolog), PTEN (phosphatase and tensin homologue) and phosphatidylinositol- 3-kinase, PI3K) mRNA relative expression and protein expression levels. Results: (1) Compared with resting group, Akt mRNA expression was significantly down-regulated and gastrocnemius Akt protein was up-regulated in astrocyte and gastrocnemius muscle in increasing intensity exercise group; (2) PTEN mRNA, (3) The protein expression of PI3K in soleus muscle increased significantly in increasing intensity exercise group, while the relative expression of PI3K mRNA in soleus muscle and gastrocnemius muscle did not change significantly. The results suggest that Akt mRNA may be down-regulated independently of PI3K and that increasing treadmill training induces the activation of this specific Akt inactivation pathway.
