目的观察饮水无机砷暴露人外周血淋巴细胞中AS3MT基因在表达与尿砷甲基化产物的关系。方法选取饮水型砷暴露者18例,采用实时定量RT-PCR技术测定研究对象的测淋巴细胞中AS3MTmRNA的表达,采用AFS-SAP10检测尿iAs,MMA,DMA浓度,应用Pearson相关法分析AS3MTmRNA表达与MMA%,DMA%,PMI,SMI的关系。结果AS3MTmRNA表达与MMA%相关有统计学意义(r=0.485,P=0.041),与PMI相关有统计学意义(r=0.476,P=0.046);但与DMA%相关无统计学意义(r=0.187,P=0.459),与SMI相关无统计学意义(r=0.210,P=0.404)。结论人外周血淋巴细胞AS3MTmRNA表达对无机砷在体内的甲基化过重中有重要的作用。
Objective To observe the relationship between AS3MT gene expression and urinary arsenic methylation in human peripheral blood lymphocytes exposed to drinking arsenic in drinking water. Methods 18 cases of drinking arsenic exposure were selected. The expression of AS3MT mRNA in lymphocytes was detected by real-time quantitative RT-PCR. The concentrations of iAs, MMA and DMA in urine were detected by AFS-SAP10. The expression of AS3MT mRNA was analyzed by Pearson correlation MMA%, DMA%, PMI, SMI relationship. Results There was a significant correlation between the expression of AS3MT mRNA and MMA% (r = 0.485, P = 0.041) and PMI (r = 0.476, P = 0.046) 0.187, P = 0.459). There was no significant correlation with SMI (r = 0.210, P = 0.404). Conclusion The expression of AS3MT mRNA in human peripheral blood lymphocytes plays an important role in the hypermethylation of inorganic arsenic in vivo.
