目的:研究大鼠肾脏缺血再灌注损伤(IRI)后水通道蛋白-2(AQP-2)的表达变化与肾功能变化的关系。方法:建立大鼠左侧肾脏缺血再灌注损伤模型,于IRI不同的时间点处死大鼠,留取尿液、血清、肾脏标本行尿常规、肾功能、肾脏病理形态学及AQP-2的免疫组化、RT-PCR检测。结果:大鼠肾脏IRI后出现尿量增多、尿渗透压降低,血肌苷、尿素氮升高症状;HE染色示肾小管上皮细胞肿胀、坏死、脱落;AQP-2表达量及其mRNA含量均较对照组减少,这些变化于IRI 3d时最低,至IRI 7d时恢复正常。结论:肾脏IRI后AQP-2表达的减少可能是急性肾衰竭时尿量增多、尿渗透压降低的重要因素之一。
AIM: To investigate the relationship between changes of aquaporin-2 (AQP-2) and renal function after renal ischemia-reperfusion injury (IRI) in rats. Methods: The model of renal ischemia-reperfusion injury was established in rats. Rats were sacrificed at different time points of IRI. Urine, serum and kidney samples were collected for urinary routine, renal function, renal pathomorphology and AQP-2 Immunohistochemistry, RT-PCR detection. Results: After IRI, the urine volume increased, the urine osmolality decreased, and the serum creatinine and urea nitrogen increased. The renal tubular epithelial cells were swollen, necrotic and exfoliated. The expression of AQP-2 mRNA and mRNA Compared with the control group, these changes were the lowest at IRI 3d and returned to normal at 7th day after IRI. Conclusions: The decrease of AQP-2 expression after renal IRI may be one of the important factors in the increase of urine volume and the decrease of urine osmolality in acute renal failure.
