目的:研究冬凌草甲素动态积累变化关键期培养物显微、超微结构的变化,探讨培养物组织结构与冬凌草甲素积累的相关性。方法:采用高效液相色谱法研究了离体培养过程中培养物内冬凌草甲素积累的动态变化规律。在此基础之上,采用石蜡切片、透射电镜方法研究了培养物组织结构与冬凌草甲素积累的关系。结果:离体培养过程中培养物中冬凌草甲素积累表现出有-无-有的规律性,随着叶片外植体的脱分化,培养物内叶绿体数目逐渐减少,冬凌草甲素的含量也随之逐渐降低,当冬凌草甲素再次积累时,培养物内叶绿体数目再次增多且出现了维管组织的分化。结论:冬凌草甲素积累合成与培养物叶绿体数目的多少及内部维管系统的分化有着紧密的联系。
OBJECTIVE: To study the changes of microscopic and ultrastructural changes in the critical period of dynamic changes of oridonin, and to explore the correlation between the tissue structure and the accumulation of oridonin. Methods: The dynamic changes of oridonin accumulation in culture during in vitro culture were studied by high performance liquid chromatography (HPLC). On this basis, the relationship between the tissue structure and the accumulation of oridonin was studied by paraffin section and transmission electron microscopy. RESULTS: The accumulation of oridonin in the cultures during in vitro culture showed a regular pattern. With the dedifferentiation of leaf explants, the number of chloroplasts in the culture gradually decreased. The numbers of oridonin The content of chlorophyll decreased gradually. When the oridonin accumulated again, the number of chloroplasts in the culture increased again and the differentiation of vascular tissues appeared. Conclusion: The accumulation and synthesis of oridonin A is closely related to the number of chloroplasts in culture and the differentiation of internal vascular system.
