目的观察2型糖尿病大鼠肾组织细胞周期蛋白依赖激酶9(Cdk9)和细胞周期蛋白(cyclin )T1蛋白的表达及小檗碱的影响。方法小剂量注射链佐星(链脲菌素)加高糖高脂饲料喂养16周建立2型糖尿病大鼠模型,随后16周每天分别给予小檗碱75,150及300 mg·kg-1、非诺贝特100 mg·kg-1和罗格列酮4 mg·kg-1。处死大鼠后用HE染色检查肾组织结构和免疫组化技术检测Cdk9和cyclin T1的表达。结果糖尿病模型大鼠肾小球体积增大,部分系膜细胞增生和系膜区扩张,肾小球及肾小管基底膜增厚,肾重/体重比值增加;150及300 mg·kg-1小檗碱和罗格列酮能部分改善糖尿病性肾组织病变和降低肾重/体重比值。150及300 mg·kg-1小檗碱和罗格列酮都能明显恢复糖尿病肾组织中降低了的Cdk9和cyclin T1表达至接近正常水平。结论小檗碱通过调控肾组织Cdk9和cyclin T1的表达能部分改善实验性糖尿病大鼠肾组织病变。
Objective To observe the expression of cyclin-dependent kinase 9 (Cdk9) and cyclin T1 protein in renal tissue of type 2 diabetic rats and the effect of berberine. METHODS: Rat models of type 2 diabetes were established by low-dose injection of streptozotocin (streptozotocin) plus high-sugar high-fat diet for 16 weeks. Then, berberine 75, 150 and 300 mg·kg-1 were given daily for 16 weeks. Beit 100 mg·kg-1 and rosiglitazone 4 mg·kg-1. The rats were sacrificed and HE staining was used to examine the structure of the kidneys and immunohistochemistry to detect the expression of Cdk9 and cyclin T1. RESULTS: Diabetic model rats had enlarged glomerular volume, partial mesangial cell proliferation and mesangial expansion, glomerular and tubular basement membrane thickening, and increased renal weight/body weight ratio; 150 and 300 mg kg-1 Both scopolamine and rosiglitazone can partially improve diabetic renal pathological changes and reduce the kidney weight/body weight ratio. Both 150 and 300 mg·kg-1 berberine and rosiglitazone significantly restored the decreased expression of Cdk9 and cyclin T1 in the diabetic kidney tissue to near normal levels. Conclusion Berberine can partly improve the renal pathological changes in experimental diabetic rats by regulating the expression of Cdk9 and cyclin T1 in renal tissue.
