目的探讨金欣口服液治疗呼吸道合胞病毒(RSV)肺炎的可能作用机制。方法将48只BALB/c小鼠随机分为正常组、RSV肺炎模型组、金欣口服液组,每组16只。RSV肺炎模型组和金欣口服液组小鼠用RSV悬液滴鼻感染小鼠建立RSV肺炎模型。金欣口服液组予金欣口服液27.6 g/(kg·d)灌胃,正常组和RSV肺炎模型组小鼠给予等量的生理盐水灌胃。连续灌胃至第4天每组处死5只小鼠进行病理学检查,其他小鼠于第6天分别收集尿液,运用GC-MS检测尿液中所含有的代谢物及其相对含量的变化。结果RSV肺炎模型组小鼠肺脏可见间质多灶性炎性浸润,肺泡壁轻度充血,明显增厚。金欣口服液组小鼠肺部炎症较RSV肺炎模型组小鼠肺部炎症明显减轻。RSV肺炎小鼠的尿液中共测得38种代谢产物,其中乳酸、甘氨酸、棕榈酸、硬脂酸为差异性代谢产物,RSV肺炎模型组乳酸、甘氨酸、硬脂酸、棕榈酸相对含量明显高于正常组和金欣口服液组(P<0.05)。结论金欣口服液可能通过调节体内能量代谢、氨基酸代谢、脂肪酸代谢治疗RSV肺炎。
Objective To investigate the possible mechanism of Jinxin oral liquid in the treatment of respiratory syncytial virus (RSV) pneumonia. Methods 48 BALB / c mice were randomly divided into normal group, RSV pneumonia model group and Jinxin oral liquid group, 16 in each group. RSV pneumonia model group and Jinxin oral liquid group mice were intranasally infected with RSV suspension to establish RSV pneumonia model. Jinxin oral solution group Jinxin oral solution 27.6 g / (kg · d) gavage, normal group and RSV pneumonia model mice were given the same amount of saline gavage. Five rats were sacrificed on the fourth day after operation. Urine was collected on the sixth day in other mice. The metabolites and their relative contents in urine were detected by GC-MS . Results In the RSV pneumonia model group, interstitial multifocal inflammatory infiltration was observed in the lungs of mice. The alveolar wall was mildly hyperemic and obviously thickened. Inflammation in the lung of mice in Jinxin oral liquid group was significantly reduced than that of mice in RSV pneumonia model. A total of 38 metabolites were detected in the urine of RSV pneumonia mice. Among them, lactic acid, glycine, palmitic acid and stearic acid were the differential metabolites. The relative contents of lactic acid, glycine, stearic acid and palmitic acid in RSV pneumonia model group were significantly higher In the normal group and Jinxin oral liquid group (P <0.05). Conclusion Jinxin Oral Liquid may treat RSV pneumonia by regulating energy metabolism, amino acid metabolism and fatty acid metabolism in vivo.
