以TGF-β1在体外作用于人急性早幼粒白血病细胞株HL-60细胞,通过免疫细胞化学染色等实验方法,观察TGF-β1对HL-60细胞株凋亡基因表达的影响。结果发现1μg/L的TGF-β1体外作用24h后HL-60细胞中Bcl-2、C-myc基因的灰度值由27.80±3.17和20.80±4.37下调至30.56±2.57和23.42±4.45;作用72h后则分别下调至35.01±1.34和34.61±1.61。结果说明TGF-β1对人急性早幼粒白血病细胞株的凋亡基因有显著下调作用,这种作用随时间的延长更加明显。
The effect of TGF-β1 on human acute myeloid leukemia cell line HL-60 cells was studied in vitro. The effect of TGF-β1 on apoptosis gene expression in HL-60 cell line was observed by immunocytochemical staining. The results showed that the gray levels of Bcl-2 and C-myc genes in HL-60 cells were downregulated from 27.80±3.17 and 20.80±4.37 to 30 after 1 μg/L of TGF-β1 in vitro. 56±2.57 and 23.42±4.45; after 72 h, they were down-regulated to 35.01±1.34 and 34.61±1.61, respectively. The results showed that TGF-β1 had a significant down-regulation of apoptosis genes in human acute promyelocytic leukemia cell lines, and this effect was more pronounced with time.
