以西藏帕里产粗壮秦艽为研究对象,采用硅胶柱、Sephadex LH-20凝胶柱等手段进行化学成分的分离纯化,通过波谱数据(MS,~1H-NMR,~(13)C-NMR)及标准品对照进行结构鉴定;进一步利用HPLC法测定该植物不同器官(根、茎、叶、花)中几种成分的含量并比较含量差异.从西藏帕里产粗壮秦艽中分离得到5个化合物,分别鉴定为栎瘿酸(1)、谷甾醇(2)、龙胆苦苷(3)、马钱苷酸(4)、獐牙菜苦苷(5),其中环烯醚萜苷类化合物马钱苷酸、獐牙菜苦苷、龙胆苦苷、獐牙菜苷的含量较高,且在4种器官中含量差异明显.本研究首次从西藏帕里产粗壮秦艽中分离得到化合物1-5,其中环烯醚萜苷类含量丰富,在4种器官中马钱苷酸和龙胆苦苷总含量均达到2015年版药典标准,因此根、茎、叶与花均具有同等重要的开发价值.
In order to study the chemical constituents of Gentiana macrophylla from Pall in Tibet, the chemical constituents were isolated and purified by silica gel column and Sephadex LH-20 gel column. The spectral data (MS, ~ 1H-NMR and ~ (13) C-NMR) And the standard control were further identified by HPLC.The contents of several components in different organs (roots, stems, leaves, flowers) of this plant were further determined by HPLC and the content differences were compared.The results showed that 5 compounds (1), sitosterol (2), gentiopicroside (3), stilbenzoic acid (4), swertia glycoside (5), respectively. Among them, iridoid glycosides Maltosinic acid, swertia bitter glycosides, gentiopicroside, swertia glycosides content is high, and the contents of the four organs were significantly different.In this study, for the first time from the Palyan caddity in Tibet to isolate compound 1 -5, of which iridoid glycosides are abundant, and the total contents of ramification and gentiopicroside in all four organs reached the standard of Pharmacopoeia in 2015. Therefore, root, stem, leaf and flower all have the same important development value.
