目的分析吉林省延边朝鲜族自治州(延边州)流行的肠道病毒71型(EnterovirusType71,EV71)分离株的基因特征。方法对2009~2010年延边州手足口病(Hand,Footand Mouth Disease;HFMD)来源的5株EV71分离株进行逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)扩增VP1编码区,测序并使用Bioedit和Maga4生物信息学软件进行基因特征分析。结果延边州5株EV71分离株均为C4a基因亚型,其VP1编码区核苷酸序列具有较高的同源性(99.3%~100%),在基因树上形成紧密的一簇。延边州的5株分离株与安徽省阜阳市2008年流行株具有最近的基因亲缘关系,核苷酸同源性高达99.3%~99.4%。结论延边州2009~2010年EV71分离株均属于C4a亚型,至少有一个EV71传播链在延边州持续传播,引起延边州HFMD的流行。需要加强EV71分子流行病学监测,掌握本地流行的EV71基因型数据,并及时发现输入基因型。
Objective To analyze the genetic characteristics of Enterovirus Type 71 (EV71) isolates prevalent in Yanbian Korean Autonomous Prefecture (Yanbian Prefecture) in Jilin Province. Methods Five EV71 isolates from Hand, Foot and Mouth Disease (HFMD) from 2009 to 2010 were amplified by RT-PCR Coding region, sequenced and gene characterization was performed using Bioedit and Maga4 bioinformatics software. Results All of the five EV71 isolates in Yanbian were C4a genotypes. The nucleotide sequence of the VP1 coding region was highly homologous (99.3% -100%), forming a close cluster in the gene tree. The five isolates in Yanbian and the epidemic strains in Fuyang in Anhui Province in 2008 have the closest genetic relationship with nucleotide homology of 99.3% -99.4%. Conclusion EV71 isolates in Yanbian Prefecture from 2009 to 2010 belonged to C4a subtypes. At least one EV71 transmission chain continued to spread in Yanbian prefecture, causing epidemic of HFMD in Yanbian prefecture. It is necessary to strengthen the molecular epidemiological surveillance of EV71, master the EV71 genotype data of the local epidemic, and find the input genotype in time.
