目的:研究凝血酶在体外对非小细胞肺癌细胞(A549)增殖、化疗药物顺铂疗效的影响及其机理。方法:非小细胞肺癌细胞株A549与不同浓度的凝血酶(0.05-3U/ml)共培养,按不同分组加入顺铂(30mg/ml)和水蛭素(0.05-10U/ml)。在不同的时间,采用MTT比色法检测细胞生长活性,细胞增殖程度;采用流式细胞术检测凝血酶对顺铂杀伤肿瘤细胞作用的影响。结果:凝血酶在一定浓度范围内(0.05-1.5 U/ml)可促进A549增殖,72 h后增殖效应明显;在增殖浓度范围内可抑制顺铂对肿瘤细胞A549的凋亡作用。结论:凝血酶在一定浓度范围内诱导A549细胞增殖,并减弱化疗药物顺铂对该细胞的杀伤作用。
Objective: To study the effect and mechanism of thrombin on the proliferation of non-small cell lung cancer cells (A549) and the chemotherapeutic drug cisplatin in vitro. Methods: Non-small cell lung cancer cell line A549 was co-cultured with different concentrations of thrombin (0.05-3U / ml), and cisplatin (30mg / ml) and hirudin (0.05-10U / ml) were added in different groups. At different times, MTT colorimetric assay was used to detect cell growth activity and cell proliferation. Flow cytometry was used to detect the effect of thrombin on cisplatin-induced tumor cell killing. Results: Thrombin could promote the proliferation of A549 in a certain concentration range (0.05-1.5 U / ml), and the proliferative effect was obvious after 72 hours. It could inhibit the apoptosis of A549 cells induced by cisplatin in the concentration range. Conclusion: Thrombin induces the proliferation of A549 cells in a certain concentration range and attenuates the killing effect of cisplatin on the cells.
