COP1(constitutive photomorphogenesis 1)是光形态建成的一个抑制因子。本研究采用快速扩增cDNA末端(rapid-amplification of cDNA ends,RACE)技术克隆得到了甘薯COP1的cDNA序列,并通过实时荧光定量PCR检测其在甘薯不同组织部位的表达情况。结果表明,获得的COP1的cDNA全长为2 224 bp,含有完整的开放阅读框(2 016 bp),编码671个氨基酸,并命名为ibCOP1(Gen Bank登录号:KT749985)。序列对比结果显示,甘薯ibCOP1与牵牛花的COP1氨基酸序列相似性高达96%。实时荧光定量PCR结果显示,ibCOP1在甘薯根、茎、叶和花中都有表达,在成熟紫叶中表达量最高,根中表达量最低。本研究首次克隆甘薯COP1基因,为进一步研究其生理功能奠定基础。
COP1 (constitutive photomorphogenesis 1) is a suppressor of light morphogenesis. In this study, the cDNA sequence of sweet potato COP1 was cloned by rapid amplification of cDNA ends (RACE). The expression of COP1 in sweet potato was detected by real-time fluorescence quantitative PCR. The results showed that the full length cDNA of COP1 was 2 224 bp in length and contained a complete open reading frame (2 016 bp) encoding 671 amino acids and named ibCOP1 (Gen Bank accession number: KT749985). Sequence comparison showed that the amino acid sequence similarity of COP1 between sweet potato ibCOP1 and morning glory was as high as 96%. Real-time PCR results showed that ibCOP1 was expressed in roots, stems, leaves and flowers of sweet potato, and highest in mature purple leaves and lowest in roots. This study cloned the first COP1 gene of sweet potato for further study of its physiological function.
