目的探讨华支睾吸虫颗粒蛋白(CsGRN)在虫体和宿主中的表达。方法使用荧光定量PCR(qRT-PCR)检测华支睾吸虫成虫、囊蚴和虫卵及猫的肝脏、胆管组织和全血CsGRN基因mRNA表达水平;免疫组化检测CsGRN在成虫及宿主猫、人和实验感染的Balb/c小鼠肝脏中的定位。结果 CsGRN基因mRNA在虫体的各阶段都有表达,在感染的猫肝和胆管组织中高表达;CsGRN主要定位在成虫的睾丸、外膜和虫卵,在宿主人、猫和小鼠中主要分布在胆管上皮和肝细胞。结论 CsGRN可能参与虫体的生长发育,它作为分泌排泄抗原之一可能损害肝胆管组织,有进一步研究价值。
Objective To investigate the expression of CsGRN in worms and hosts. Methods Quantitative PCR (qRT-PCR) was used to detect the mRNA expression of CsGRN in adults, metacercariae and ovum and the liver, bile duct tissue and whole blood of cats. Immunohistochemistry was used to detect the expression of CsGRN in adult and host cats, And the location in the liver of experimentally infected Balb / c mice. Results CsGRN mRNA was expressed in all stages of the worms and highly expressed in the infected cats liver and bile duct tissues. CsGRN mainly located in the testis, adventitia and eggs of adult worms, and mainly distributed in host human, cat and mouse In the bile duct epithelium and liver cells. Conclusion CsGRN may be involved in the growth and development of parasites. As one of the secreted and excreted antigens, CsGRN may impair the hepatobiliary and biliary tissues, which is of further research value.
