目的探讨不同浓度ERK抑制剂PD98059体外对泡状棘球蚴原头节生长的作用及形态学影响。方法将体外培养的泡状棘球蚴分别加入6.25、12.5、25、50μmol/L PD98059中体外孵育,用0.1%伊红染色后在倒致显微镜下观察原头节形态并检测活力,试验重复3次;于透射电子显微镜(TEM)下观察PD98059作用后原头节超微结构变化;采用Caspase-3活性检测试剂盒检测原头节caspase-3酶活性。结果 DMEM/DMEM+DMSO培养组原头节活力较开始时无明显变化。50μmol/L PD98059作用48h后,原头节活力下降为(55.16±0.212)%,作用第14d,无存活的原头节。低浓度组对原头节的杀伤作用较高浓度组弱。TEM观察PD98059处理后的原头节微毛及糖萼出现部分缺损,合胞体带内出现少量小的空泡,且随用药浓度的增大,空泡增大增多并出现脂滴。ELISA检测显示,经PD98059处理的原头节caspase-3酶活性较正常对照组增强。结论 ERK抑制剂PD98059在体外有抗泡状棘球蚴作用。
Objective To investigate the effect of different concentrations of PD98059, an ERK inhibitor, on the growth of prothallial cells of Echinococcus granulosus and its morphological effects. Conclusion The ERK inhibitor PD98059 has anti-bubble-form hydatid cyst in vitro.
