为了研究大孔吸附树脂对山丹鳞茎中皂苷静态吸附和解吸性能,筛选出纯化效果较好的树脂,试验采用静态吸附筛选树脂和动态吸附确定工艺参数的方法来研究大孔吸附树脂对山丹鳞茎中皂苷的分离纯化工艺。结果表明:D101大孔吸附树脂对山丹鳞茎中皂苷的纯化效果较好,在上样原液浓度为6 mg/m L、pH值为8的条件下进行吸附,解吸条件为乙醇浓度70%、洗脱流速0.5 BV/h。在此条件下对山丹鳞茎中皂苷进行纯化,皂苷纯度提高了4.35倍。说明D101大孔吸附树脂对山丹鳞茎皂苷有较好的富集、纯化效果。
