目的研究低剂量辐射预先照射以及随后大剂量照射后小鼠胸腺细胞T细胞受体(TCR)、CD3、CD4和CD8分子表达的变化。方法TCR、CD3表达采用间接荧光流式细胞术检测,CD4、CD8表达采用双参数直接免疫荧光流式细胞术检测。结果实验结果表明:单纯15GyX射线全身照射后TCR,CD3阳性细胞数以及CD4-D8-,CD4+CD8+,CD4+CD8-和CD4-CD8+细胞数明显减少,当15GyX射线全身照射前6小时预先照射0075Gy时,可明显减轻其后15Gy照射对TCR+,CD3+,CD4+CD8+,CD4+CD8-和CD4-CD8+的损伤作用。表现为各亚组细胞数显著高于单纯15Gy照射组。CD4-CD8-亚组的细胞数无明显变化。结论0075GyX射线全身照射能够诱导胸腺细胞TCR+,CD3+,CD4+CD8+,CD4+CD8-和CD4-CD8+亚组细胞的适应性反应。
Objective To study the changes of T cell receptor (TCR), CD3, CD4 and CD8 expression in mouse thymocytes after pretreatment with low dose radiation and subsequent high dose irradiation. Methods TCR and CD3 expression were detected by indirect fluorescent flow cytometry. The expression of CD4 and CD8 were detected by two-parameter direct immunofluorescence flow cytometry. Results The experimental results showed that the number of TCR, CD3 positive cells and the number of CD4-D8-, CD4 + CD8 +, CD4 + CD8- and CD4-CD8 + cells decreased significantly after irradiation with 1.5Gy X-ray and were pre-irradiated 6 hours before the whole body irradiation At 0075 Gy, the injury of TCR +, CD3 +, CD4 + CD8 +, CD4 + CD8- and CD4-CD8 + after 15 Gy irradiation was significantly alleviated. The performance of each subgroup of cells was significantly higher than the simple 1 5Gy irradiation group. There was no significant change in the number of CD4-CD8-subgroups. Conclusion 0075 Gy X-rays can induce the adaptive response of TCR +, CD3 +, CD4 + CD8 +, CD4 + CD8- and CD4-CD8 + subsets in thymocytes.
