随着透射电子显微镜(TEM)在单细胞超微形态学研究方面的广泛应用,对于收集保存悬浮游离细胞(如培养细胞悬液、临床上细胞学检查的送检体液等),就显得非常重要。以往制备悬浮游离细胞电镜样品,通常采用从提取细胞到环氧树脂包埋步步离心的方法。由于这种收集方法离心次数多(十几次),细胞丢失亦多。致使包埋出的样品显得稀疏不集中,细胞切片机遇性小,给研究工作带来很多不便。为此,本文设计使用琼脂糖凝胶作预包埋,使细胞在锇酸固定前就再不呈游离状态,控制了细胞的丢失现象。经工作使用证明,该方法收
