目的:观察活血熄风方对局灶性脑缺血大鼠脑组织中Caspase-3蛋白及mRNA表达的影响。方法:光化学法建立局灶性脑缺血大鼠模型,免疫荧光与激光共聚焦技术检测大脑缺血皮层Caspase-3表达荧光强度和阳性细胞计数,逆转录聚合酶链式反应(RT-PCR)法检测Caspase-3mRNA表达。结果:与模型组比较,活血熄风方治疗组的Caspase-3蛋白及mRNA表达明显降低(P<0.05或P<0.01)。结论:活血熄风方对大鼠局灶性脑缺血损伤具有保护作用,其作用机制可能与抑制Caspase-3蛋白及mRNA表达有关。
Objective: To observe the effect of Huoxue Xifeng Decoction on the expression of Caspase-3 protein and mRNA in rat brain after focal cerebral ischemia. METHODS: A rat model of focal cerebral ischemia was established by photochemical method. Immunofluorescence and confocal laser scanning was used to detect the expression of Caspase-3 in cerebral ischemic cortex and the positive cell counts. RT-PCR was used. The method detects Caspase-3 mRNA expression. RESULTS: Compared with the model group, the expression of Caspase-3 protein and mRNA was significantly lower in the Huoxue Xifeng Recipe group (P<0.05 or P<0.01). Conclusion: Huoxue Xifeng Decoction has protective effects on focal cerebral ischemic injury in rats, and its mechanism may be related to the inhibition of Caspase-3 protein and mRNA expression.
