目的比较葡萄汁与单体白黎芦醇对镉染毒小鼠的抗氧化损伤作用和促排镉作用及其作用机制。方法将32只5周龄清洁级昆明小鼠随机分为4组,分别为空白对照(双蒸水)组、镉染毒(50 mg/L)组、葡萄汁(含白藜芦醇65μg/ml)+镉染毒(50 mg/L)组、白藜芦醇(65μg/ml)+镉染毒(50 mg/L)组,每组8只,雌雄各半。氯化镉溶液采用自由饮用方式进行染毒;葡萄汁和白藜芦醇溶液采用灌胃方式进行染毒,染毒容量为0.02 ml/g,每天1次,连续染毒4周。染毒结束后,测定肝、肾组织中的镉含量,血清中尿素氮(BUN)的含量及谷丙转氨酶(ALT)的活力,肝、肾组织和血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活力和丙二醛(MDA)的含量。结果与镉染毒组相比,葡萄汁+镉染毒组和白藜芦醇+镉染毒组小鼠肝脏、肾脏中的镉含量均下降;血清中SOD、GSH-Px活力升高,ALT活力和BUN、MDA含量下降;肝、肾组织中SOD、GSH-Px活力升高,MDA含量下降,差异有统计学意义(P<0.01)。与白藜芦醇+镉染毒组比较,葡萄汁+镉染毒组小鼠肝脏、肾脏中的镉含量下降;血清中SOD、GSH-Px活力升高,ALT活力和BUN、MDA含量下降;肝组织中SOD活力升高,MDA含量下降;肾组织中SOD、GSH-Px活力升高,MDA含量下降,差异均有统计学意义(P<0.01)。结论葡萄汁对镉中毒小鼠排镉和肝、肾损伤的抗氧化作用强均于白黎芦醇。
OBJECTIVE To compare the anti-oxidative and anti-oxidative effects of grape juice and monomeric resveratrol on cadmium-exposed mice and its mechanism. Methods Totally 32 clean Kunming mice of 5 weeks age were randomly divided into 4 groups: blank control group (double distilled water), cadmium exposure group (50 mg / L), grape juice (containing resveratrol 65 μg / ml) + cadmium exposure group (50 mg / L), resveratrol (65 μg / ml) + cadmium exposure group (50 mg / L), with 8 mice in each group. Results Compared with the cadmium exposure group, the cadmium levels in the liver and kidney of grape juice + cadmium poisoning group and resveratrol + cadmium poisoning group were decreased; the activities of SOD and GSH-Px in serum were increased, and the levels of ALT The activities of SOD and GSH-Px in liver and kidney were increased, and the contents of MDA and MDA were decreased (P <0.01). Compared with the resveratrol + cadmium group, the content of cadmium in the liver and kidney of the grape juice + cadmium group decreased; the activity of SOD and GSH-Px in the serum increased, the activity of ALT and the content of BUN and MDA decreased; The activities of SOD and GSH-Px in the liver tissue increased and the content of MDA decreased in the liver tissue (P <0.01). Conclusion grape juice on cadmium poisoning row of cadmium and liver and kidney damage in the antioxidant activity are stronger in resveratrol.
