[目的]探讨缺氧诱导因子-1α(HIF-1α)在子痫前期胎盘组织中的表达及意义。[方法]选取2008年9月~2009年12月在某院分娩病例56例。分为:对照组:正常孕妇30例;子痫前期组26例,其中轻度12例、重度14例。所有病例均排除内、外科合并症及其他产科并发症,均为单胎。应用免疫组织化学方法及Western blot法,比较各组胎盘组织HIF-1α的表达。[结果]HIF-1α的表达主要位于胎盘滋养细胞胞核;与正常分娩妇女胎盘组织相比较,子痫前期患者胎盘组织中HIF-1α染色程度及表达水平明显升高(P﹤0.01);且随着病情的加重,染色程度、表达水平亦呈上升趋势;各组间表达差异有统计学意义(P﹤0.01)。[结论]HIF-1α在子痫前期患者胎盘组织中的高表达,从蛋白水平进一步证实缺氧为子痫前期发病机制之一。
[Objective] To investigate the expression and significance of hypoxia inducible factor-1α (HIF-1α) in placenta of preeclampsia. [Method] Choose 56 cases of childbirth in a hospital from September 2008 to December 2009. Divided into: control group: 30 cases of normal pregnant women; preeclampsia group of 26 cases, of which 12 cases were mild, severe in 14 cases. All cases were excluded, surgical complications and other complications of obstetrics, are singleton. Immunohistochemistry and Western blot were used to detect the expression of HIF-1α in placenta of each group. [Results] The expression of HIF-1α was mainly located in the nucleus of placental trophoblasts. Compared with the normal childbirth women, the level of HIF-1α staining and the expression of placental tissue in preeclampsia were significantly increased (P <0.01). With the aggravation of the disease, the staining degree and expression level also showed an upward trend. The differences among the groups were statistically significant (P <0.01). [Conclusion] HIF-1α is highly expressed in the placenta of preeclampsia patients, further confirming the hypoxia as one of the pathogenesis of preeclampsia from the protein level.
