采用免疫组织化学ABC法对30耳中耳胆脂瘤、11耳健康外耳道皮肤进行表皮生长因子受体(EGFR)定位,用酶联免疫电转移印迹技术(EITB)检测8耳中耳胆脂瘤、4耳健康外耳道皮肤中EGFR。结果:130耳中,29耳上皮、24耳成纤维细胞、单个核细胞及血管内皮细胞显示EGFR阳性,免疫着色主要定位于细胞膜及细胞浆,且从基底细胞层到角质层逐渐减弱;外耳道健康皮肤仅散在基底细胞和棘细胞及6耳血管内皮细胞呈弱阳性。28耳胆脂瘤标本中有6耳在170KD处出现阳性蛋白带,所有外耳道健康皮肤未见任何免疫反应阳性产物。对EGFR在中耳胆脂瘤中的可能作用进行了探讨
Epidermal growth factor receptor (EGFR) was localized in 30 ears of middle ear cholesteatoma and 11 ears of healthy external auditory meatus by immunohistochemical ABC method. Eighteen cases of middle ear cholesteatoma were detected by enzyme-linked immunosorbent assay (EITB) , 4 ears healthy external auditory canal skin EGFR. Results: 130 ears, 29 ears epithelium, 24 ears fibroblasts, mononuclear cells and vascular endothelial cells showed positive EGFR. Immunofluorescence mainly located in the cell membrane and cytoplasm, and gradually decreased from the basal cell layer to the stratum corneum. External auditory meatal healthy skin scattered only in basal cells and spine cells and 6 ear vascular endothelial cells were weakly positive. 2 8 ear cholesteatoma samples in 6 ears at 170KD at a positive protein band, all healthy skin outside the ear canal did not see any immunopositive positive product. The possible role of EGFR in middle ear cholesteatoma was explored
