目的通过检测子宫内膜异位症患者血清及腹腔液中明胶酶(MMP-2、MMP-9)的活性,探讨其在子宫内膜异位症发病中的作用。方法2004年6月至2005年6月对吉林大学第二医院妇科60例子宫内膜异位症患者采用含SDS-明胶的聚丙烯酰胺凝胶电泳为基础的酶谱法,检测血清和腹腔液MMP-2、MMP-9活性,其中Ⅰ~Ⅱ期22例,Ⅲ~Ⅳ期38例,良性卵巢囊肿患者20例为对照组。结果子宫内膜异位症患者血清和腹腔液中MMP-2、MMP-9明显高于对照组(P<0·05)。随着子宫内膜异位症(EMs)分期的增加,其血清和腹腔液中MMP-2表达量呈上升趋势,且血清中EMsⅢ~Ⅳ期组MMP-2酶活性比对照组增高2倍(P<0·01)。结论酶谱法是诊断子宫内膜异位症的有效手段,内异症患者明胶酶MMP-2、MMP-9的表达水平增高,其在子宫内膜异位症的发生发展中具有重要作用。
Objective To explore the role of endometriosis in the pathogenesis of endometriosis by detecting the activity of gelatinase (MMP-2, MMP-9) in serum and peritoneal fluid of patients with endometriosis. Methods From June 2004 to June 2005, 60 patients with endometriosis in Department of Gynecology, the Second Hospital of Jilin University were enrolled in this study. SDS-gelatin-based polyacrylamide gel electrophoresis was used to detect serum and peritoneal fluid MMP-2 and MMP-9 activity, of which 22 cases were stage Ⅰ-Ⅱ, 38 cases were stage Ⅲ-Ⅳ, and 20 cases of benign ovarian cyst were control group. Results Serum and peritoneal fluid MMP-2 and MMP-9 in patients with endometriosis were significantly higher than those in the control group (P <0.05). The expression of MMP-2 in serum and peritoneal fluid increased with the increase of staging of endometriosis (EMs), and the activity of MMP-2 in serum of EMs Ⅲ ~ Ⅳ group increased by 2 folds P <0.01). Conclusion Zymography is an effective method to diagnose endometriosis. The expression of MMP-2 and MMP-9 in patients with endometriosis is increased, which plays an important role in the development of endometriosis.
