目的探讨细胞周期相关激酶(CCRK)在大肠癌组织中的表达及其与大肠癌临床病理特征之间的关系。方法选取50例经病理学确诊的原发性大肠癌患者的癌组织,以配对的癌旁组织作为对照,采用RT-PCR和Western blot检测CCRK mRNA和CCRK蛋白表达的差异性。结果 CCRK蛋白在大肠癌组织中的阳性率为60.00%(30/50),癌旁结肠组织阳性率为40.00%(20/50)差异有统计学意义(χ2=5.769,P<0.05);CCRK蛋白在大肠癌组织中的相对表达量为2.69±0.87,癌旁组织为1.00±0.28,差异有统计学意义(t=5.81,P<0.05)。CCRK mRNA在大肠癌组织中的阳性率为64.00%,癌旁组织为42.00%,差异有统计学意义(χ2=4.857,P<0.05);CCRK mRNA在大肠癌组织中的相对表达量为2.45±0.92,癌旁组织为1.00±0.21,差异有统计学意义(t=4.83,P<0.05)。PT3-4期大肠癌组织CCRK mRNA阳性率为85.70%(12/14),PT1-2期大肠癌组织阳性率55.56%(20/36),差异有统计学意义(χ2=3.979,P<0.05);PN1-2期大肠癌组织CCRK mRNA阳性率为87.50%(14/16),PN0期为52.94%(18/34),差异有统计学意义(χ2=5.640,P<0.05)。结论 CCRK的高表达与大肠癌发生、发展及其TNM分期有关,检测CCRK蛋白和CCRK mRNA表达可以作为大肠癌早期诊断和预后的一项重要参考指标。
Objective To investigate the expression of cyclin-dependent kinase (CCRK) in colorectal carcinoma and its relationship with the clinicopathological features of colorectal cancer. Methods Fifty cases of pathologically diagnosed primary colorectal cancer were selected as the control. Paired paracancerous tissues were used as control. The differences of CCRK mRNA and CCRK protein expression were detected by RT-PCR and Western blot. Results The positive rate of CCRK protein in colorectal cancer was 60.00% (30/50), while the positive rate of colorectal cancer was 40.00% (20/50) (χ2 = 5.769, P <0.05) The relative expression of protein in colorectal cancer was 2.69 ± 0.87, paracancerous tissue was 1.00 ± 0.28, the difference was statistically significant (t = 5.81, P <0.05). The positive rate of CCRK mRNA in colorectal carcinoma was 64.00%, while the adjacent tissue was 42.00% (χ2 = 4.857, P <0.05). The relative expression of CCRK mRNA in colorectal carcinoma was 2.45 ± 0.92, paracancerous tissue 1.00 ± 0.21, the difference was statistically significant (t = 4.83, P <0.05). The positive rates of CCRK mRNA in PT3-4 colorectal cancer tissues were 85.70% (12/14), and those in PT1-2 colorectal cancer tissues were 55.56% (20/36) (χ2 = 3.979, P <0.05) ). The positive rate of CCRK mRNA in PN1-2 colorectal cancer tissues was 87.50% (14/16), PN0 stage was 52.94% (18/34), the difference was statistically significant (χ2 = 5.640, P <0.05). Conclusion The high expression of CCRK is associated with the occurrence, development and TNM staging of colorectal cancer. Detection of CCRK protein and CCRK mRNA expression may be an important reference for the early diagnosis and prognosis of colorectal cancer.
