该实验通过建立大鼠下腔静脉血栓模型和小鼠急性肺血栓模型,考察鬼箭锦鸡儿醇提物(TE)的抗血栓作用。通过考察TE的2个主要部分TE-1和TE-2对小鼠急性肺血栓的作用,以及比较TE、TE-1和TE-2对出血时间和凝血时间的影响,探索其抗血栓活性部位。通过对照品对照结合液相色谱-质谱分析对提取物中所含6种主要成分及其他成分的种类或类别进行鉴定。结果表明,TE各剂量组均能够显著降低大鼠下腔静脉血栓质量(P<0.01),中、高剂量时能够提高对急性肺血栓模型小鼠的保护率(P<0.05)。TE-2在保护小鼠免于偏瘫或死亡、延长出血时间和凝血时间方面均比TE-1显示出较强活性。TE中主要含有异黄酮、紫檀素及茋类化合物,TE-2中以异黄酮和紫檀素类成分为主,TE-1中以茋类为主,提示这3类成分可能均为鬼箭锦鸡儿抗血栓的活性成分。
This experiment through the establishment of rat model of inferior vena cava thrombosis and mouse acute pulmonary thrombus model to investigate the anti-thrombotic effect of Caragana korshinskii ethanol extract (TE). By investigating the effect of TE-1 and TE-2, two major components of TE, on the acute pulmonary thrombus in mice and comparing the effects of TE, TE-1 and TE-2 on the bleeding time and clotting time, the antithrombotic active site . The types and categories of the 6 main components and other components contained in the extract were identified by reference standard and liquid chromatography-mass spectrometry. The results showed that all TE groups could significantly reduce the quality of inferior vena cava thrombus in rats (P <0.01), and the protection rate of acute pulmonary thrombus model mice at medium and high doses (P <0.05). TE-2 showed stronger activity than TE-1 in protecting mice from hemiplegia or death, prolonging bleeding time and clotting time. TE mainly contains isoflavones, red sandalwood and stilbene compounds, TE-2 in isoflavones and red sandalwood-based ingredients, TE-1 in the main class, suggesting that these three components may be Ghost Arrow Rock Carassius auratus Antithrombotic active ingredients.
