目的:观察真武汤对心力衰竭大鼠血清ST2及IL-33的影响,并探讨其作用机制。方法:以80只健康雄性Wistar大鼠为研究对象,除空白组10只外,其余70只通过冠脉结扎手术,术后结合冷水力竭式游泳,复制心力衰竭动物模型,把造模后存活的50只大鼠随机分为真武汤高剂量组、中剂量组、低剂量组、西药组和模型组5组,分别予以真武汤、卡托普利、生理盐水灌胃,疗程15 d。用酶联免疫吸附试验法(ELISA)测定大鼠血浆ST2、白细胞介素-33(IL-33)水平。结果:中药高剂量组、西药组FS值与EF值与模型组比较显著升高(P<0.01),中药高剂量组、西药组大鼠FS值、EF值与正常组比较差异无统计学意义(P>0.05);模型组ST2、IL-33水平均显著升高(P<0.01),中药高剂量组、中剂量组与西药组ST2、IL-33水平显著降低(P<0.01),西药组与中药高剂量组、中剂量组比较差异无统计学意义(P>0.05),中药高剂量组、中剂量组IL-33水平较正常组无统计学意义(P>0.05)。结论:真武汤能够明显降低心力衰竭大鼠血清ST2、IL-33水平,说明真武汤通过调控IL-33/ST2L信号通路改善心室重构,这可能是真武汤治疗心力衰竭的机制之一。
Objective: To observe the effect of Zhenwu decoction on serum ST2 and IL-33 in heart failure rats and to explore its mechanism. Methods: Totally 80 healthy male Wistar rats were selected as study object. Except for 10 rabbits in the blank group, the other 70 rabbits were subjected to coronary artery ligation, followed by cold water exhausted swimming. The animal model of heart failure was replicated and survived after modeling 50 rats were randomly divided into Zhenwu Decoction high dose group, middle dose group, low dose group, western medicine group and model group and were given Zhenwu Decoction, captopril and saline respectively for 15 days. Serum ST2 and interleukin-33 (IL-33) levels were measured by enzyme-linked immunosorbent assay (ELISA). Results: The values of FS and EF in high-dose group and western-medicine group were significantly higher than those in model group (P <0.01). There was no significant difference in FS value and EF between Chinese medicine group and western medicine group (P <0.05). The levels of ST2 and IL-33 in the model group were significantly increased (P <0.01). The levels of ST2 and IL-33 in the high dose group, middle dose group and western medicine group were significantly decreased There was no significant difference between high dose group and middle dose group (P> 0.05). The level of IL-33 in high dose group and middle dose group was not statistically significant (P> 0.05). Conclusion: Zhenwu decoction can significantly reduce serum levels of ST2 and IL-33 in heart failure rats, indicating that Zhenwu Decoction can improve ventricular remodeling through regulating IL-33 / ST2L signaling pathway, which may be one of the mechanisms of Zhenwu Decoction in treating heart failure.
