[目的]探讨丁苯酞对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的影响及机制.[方法]选取清洁级大鼠54只(雄性),建立UUO模型,随机分为3组,每组各18只:治疗组(A组)和模型组(B组)于靠近肾盂段行左输尿管结扎,A组予丁苯酞(NBP)180mg/kg/d灌胃;B组予等量生理盐水灌胃;假手术组(C组)仅分离输尿管,不行结扎,予等量生理盐水灌胃.各组在术后d2、d3、d7 取大鼠肾脏和血液样品,测定肾脏脂质过氧化物反应标记物超氧化物歧化酶(T-SOD)和丙二醛(MDA)活性.[结果]A组、B组、C组术后d3、d7 与d2 相比T-SOD水平降低,MDA水平升高(P<0.05).三组术后d7分别与术后d3相比T-SOD水平降低,MDA水平升高(P<0.05).B组术后d3、d7与A组相比T-SOD水平降低,MDA水平升高(P<0.05).A组术后d3、d7 与C组相比T-SOD水平降低,MDA水平升高(P<0.05).A组、B组术后d3、d7 分别与同组术后d2 相比Ⅰ型胶原蛋白质水平均升高(P<0.05).且B组术后d3、d7高于A组(P0.05).[结论]NBP对UUO大鼠肾间质纤维化能够起到明显的调控作用,其原因可能与调节了 MDA及T-SOD的活性具有一定的关系.“,”[Objective]ToobservetheeffectsofbutyLphthaLide(NBP)ontherenaLinterstitiaLfibrosisinrats withuniLateraLureteraLobstruction(UUO)andtoexpLorethepossibLemechanism.[Methods]AtotaLof54cLean LeveLmaLeratswereseLectedtoestabLishtheUUO modeLs.TheseratswererandomLydividedintothreegroups with18ineachgroup:groupAandBwithuniLateraLureteraLobstruction(LeftureteraLLigationincLoseproximity torenaLpeLvis)andgroupCwithoutUUO (proceduremimicascontroLgroup).RatsingroupAreceivedNBP180 mg/kg/dLavage,whiLeratsingroupBreceivedequaLdosagesaLineLavage.RatsingroupCwereonLyisoLatedthe ureterwithnoLigationbutgiveequaLsaLineLavageasweLL.RatkidneyandbLoodsampLesweretakenatpostopera-tived2,d3,andd7,andrenaLLipidperoxidereactionmarkerssuperoxidedismutase(T-SOD)andmaLondiaLdehyde (MDA)activitiesweremeasured.[ResuLts]Comparedtod2,theLeveLsofT-SODatpostoperatived3 andd7in groupA,groupBandgroupCdecreased,whiLetheLeveLofMDAincreased (P<0.05).T-SODLeveLwasde-creasedand MDALeveLwasincreasedinaLLthreegroupsatd7aftersurgery,ifcomparedtod3(P<0.05).Com-paredtogroupA,theLeveLofT-SODingroupBwasdecreased,whiLetheLeveLofMDA wasincreasedatpostop-eratived3andd7(P<0.05).ComparedwithgroupC,theLeveLofT-SOD wasdecreasedandtheLeveLofMDA wasincreasedingroupAatpostoperatived3andd7(P<0.05).TheLeveLsoftypeIcoLLagenproteiningroupA andgroupBatpostoperatived3andd7 weresignificantLyhigherthanthoseintheirmatchedgroupatpostoperative d2(P<0.05).Further,theLeveLsoftypeIcoLLagenproteiningroupBweregreaterthanthoseingroupA.The postoperatived3andd7ofgroupBatpostoperatived3andd7 werehigherthanthoseofgroupA (P0.05).[ConcLusion]NBPpLaysasignificantroLeinthereguLationofrenaLinterstitiaLfi-brosisinratsinuniLateraLureteraLobstruction,whichmaybereLatedtothereguLationoftheactivityofMDAand T-SOD.
