目的探讨Tankyrase 1和自噬与老龄大鼠阴茎勃起功能障碍的相关性。方法不同月龄(2、8、1 6和24月龄)雄性大鼠作为研究对象,神经刺激下阴茎海绵体压力和平均动脉压比值(ICP/MAP)检测用来评价阴茎勃起功能,Masson染色检测大鼠阴茎组织平滑肌细胞和胶原纤维比值(CSMC s/CF),Westernblot检测大鼠阴茎海绵体组织中Tankyrase 1、自噬标记物LC3-Ⅰ/Ⅱ表达和自噬调节蛋白Beclin 1、p70S6K和pho spho-P70S6K(Thr3 89)的表达变化。结果老龄(24月龄)大鼠组ICP/MAP比值和CSMCs/CF比值较其8龄组显著降低(P<0.01)。Western blot检测结果 ,老龄大鼠阴茎海绵体组织中Tankyrase 1、自噬标记物LC3-Ⅰ/Ⅱ及自噬调节蛋白Beclin 1的表达比较8月龄组显著降低(P<0.05)。各月龄组自噬调节蛋白p70S6K表达差异无统计学意义(P>0.05),但是,phospho-p70S6K(Thr389)/p70S6K的比值在老龄组表达显著增加,差异有统计学意义(P<0.05)。结论老龄大鼠勃起功能障碍与阴茎海绵体组织CSMCs含量降低密切相关,其发生机制可能与CSMCs中Tankyrase 1表达降低、与自噬功能减弱有关,有待于进一步研究。
Objective To investigate the correlation between Tankyrase 1, autophagy and erectile dysfunction in aged rats. Methods Male rats of different ages (2, 8, 1, 6 and 24 months old) were used as research objects. The penis erection pressure and mean arterial pressure ratio (ICP / MAP) (CSMC s / CF) were measured. The expressions of Tankyrase 1, LC3-Ⅰ / Ⅱ and autophagy regulatory proteins Beclin 1, p70S6K and Changes in pho spho-P70S6K (Thr3 89) expression. Results The ICP / MAP ratio and CSMCs / CF ratio of aged (24-month-old) rats were significantly lower than those of the 8-day old rats (P <0.01). The results of Western blot showed that the expression of Tankyrase 1, LC3-Ⅰ / Ⅱ and autophagy-regulatory protein Beclin 1 in the corpus cavernosum of aging rats were significantly lower than those of the 8-month-old rats (P <0.05). However, the ratio of phospho-p70S6K (Thr389) / p70S6K increased significantly in aging group with statistical significance (P <0.05). There was no significant difference in the expression of p70S6K in each age group (P> 0.05) . Conclusion Erectile dysfunction in aging rats is closely related to the decrease of content of CSMCs in corpus cavernosum. The mechanism may be related to the decrease of Tankyrase 1 and the decrease of autophagy in CSMCs, and remains to be further studied.
